What is SOT antisense therapy?

SOT (Supportive Oligonucleotide Therapy) antisense therapy is a treatment in which an antisense drug that is thought to be effective in suppressing CTCs is created based on the results of gene expression analysis of CTCs obtained from cancer stem cell testing , and is injected once or twice.The antisense drug works by inhibiting the synthesis of proteins required by cancer cells, suppressing their proliferation, or inducing apoptosis in cancer cells, which is expected to have an antitumor effect.

Mechanism of action of SOT antisense therapy

Cells contain genes (DNA), and proteins are synthesized from DNA as needed. DNA consists of two strands: the strand that carries genetic information is called the sense strand, and the other strand that does not carry genetic information is called the antisense strand. Protein synthesis involves copying the information on the sense strand of DNA into RNA, which is then stripped of unnecessary parts to create mRNA, which is then delivered to the ribosome for synthesis.
On the other hand, unlike viruses and other microorganisms, human cells have RNA that is made up of a single strand. Since double-stranded RNA does not exist, if double-stranded RNA is present, it will be quickly degraded by enzymes. SOT antisense therapy takes advantage of this mechanism. Specifically, by administering RNA (antisense) that can bind to the abnormal mRNA produced by cancer cells, double-stranded RNA is produced, which is then degraded by enzymes, inhibiting the synthesis of necessary proteins.
